Finding the role of herbal medicine extracts in modulating noradrenaline and serotonin in the analgesic effect of chemotherapyinduced neuropathic pain

Objectives: Cisplatin, paclitaxel and oxaliplatin are chemotherapeutic drugs that are widely used to treat cancers. However, they can also induce side effects such as peripheral neuropathy and anorexia, which limit their use. Although several drugs are used to attenuate these negative effects, no optimal treatment is available to date. Herbal medicines extracts have been used for long time to treat various types of diseases. However, their effects and their underlying mechanism of action have not been clearly studied in chemotherapy-induced side effects. Furthermore, according to literatures, when serotonin and noradrenaline are modulated appropriately, diseases can be effectively attenuated. Thus, finding herbal medicines that could properly modulate the synthesis and action of theses neurotransmitters and alleviate the chemotherapy-induced side effects are important. Methods: Different doses of chemotherapy drugs were injected to mice to induce neuropathic pain and anorexia. To treat these side effects, ginger extracts and its components (i.e., [6]-shogaol and [6]-gingerol) were injected orally or intraperitoneally. Results: Ginger extracts and its components were able to alleviate the chemotherapy-induced side effects. These effects appeared to be mediated by changes in monoamine neurotransmitters as both the serotonergic and noradrenergic system changed after chemotherapy drugs and ginger extract treatments. Conclusion: Monoamine neurotransmitters and their receptors present in the central and peripheral nervous system play an important role in chemotherapy-induced side effects. Herbal medicine extracts can be used to alleviate these side effects. More researches are needed in the future.
